EmbryotoxicityofTransgenicRiceinRatPostimplantationWhole EmbryoCulture QianyingGuoShuangjiaWangYuWangLirenWeiJunboWangYongLi Departmentofnutritionandfoodhygiene,Schoolofpublichealth,PekingUniversity,Xueyuan Road38,Beijing100191,PRChina Background:Rice(OryzasativaL,TT51),transformedwithasyntheticcry(1)Abgenefrom BacillusthuringiensisBerliner,wasthefirstgeneticallymodified(GM)plantwiththecertification bytheMinistryofAgriculture,China.However,toxicologicalstudiesontheGMricewerestillin scarce.Previousliteraturesindicateditspotentialembryotoxicity.Wholeembryoculture(WEC) couldbeaneffectivemethodforsafetyevaluationifculturemedium,containingsubstances deprivedfromGMfood,isavailable. Method:Ratembryosexplantedfromuteruswererandomlydistributedinto5groups:Transgenic Rice(TR)groupwithculturemediumcontainingtransgenicricedietaryadministration,Parental Rice(PR)groupwithculturemediumcontainingparentalricedietaryadministration,Blank Control(BC)groupwithoutanyintervention,PositivecontrolA(PCA)groupwithculture mediumcontainingBisphenolAoraladministrationandPositivecontrolB(PCB)groupwith BisphenolAbeingdirectlyaddedintotheculturemedium.Afterculturingfromday9.5today 11.5ofpregnancy,embryoswerescoredforgrowthanddifferentiationattheendoftheculture period. Results:EmbryosinTRgrouphadnosignificantlylowermorphologicalscoresorsmalleryolk sacdiameter,crown-rumplength(CRL)andcranialdiametercomparingwiththoseinBCandPR group. Meanwhile,embryosinPCAandPCBgroupbothperformedsignificantlylower developmentalparametersandmorphologicalscoresthantheotherthreegroups.
