Plasmin,oneofthemostpotentandreactiveserineproteases,isinvolvedinvariousphysiologicalprocesses,includingembryo development,thrombolysis,woundhealingandcancerprogression.Theproteolyticactivityofplasministightlyregulatedthrough activationofitsprecursor,plasminogen,onlyatspeci?ctimesandinde?nedlocalesaswellasthroughinhibitionofactiveplasmin byitsabundantnaturalinhibitors.Byexploitingtheplasminogenactivatingsystemandoverexpressingdistinctcomponentsofthe plasminogenactivationcascade,suchaspro-uPA,uPARandplasminogenreceptors,malignantcellscanenhancethegenerationof plasminwhichinturn,modi?esthetumormicroenvironmenttosustaincancerprogression.Whileplasmin-mediateddegradation andmodi?cationofextracellularmatrixproteins,releaseofgrowthfactorsandcytokinesfromthestromaaswellasactivationof severalmatrixmetalloproteinasezymogens,allhavebeenafocusofcancerresearchstudiesfordecades,theabilityofplasmin tocleavetransmembranemoleculesandtherebytogeneratefunctionallyimportantcleavedproductswhichinduceoutside-in signaltransduction,hasjustbeguntoreceivesu?cientattention.Herein,wehighlightthisrelativelyunderstudied,butimportant functionoftheplasminenzymeasitisgenerateddenovoattheinterfacebetweencross-talkingcancerandhostcells. 1.Introduction actual,cleavage-executingenzyme,plasm
