A wide variety of cardiopulmonary and systemic diseases are known to lead to pulmonary hypertension (PH). A number of signalingpathwayshavebeenimplicatedinPH;however,theprecisemechanism/sleadingtoPHisnotyetclearlyunderstood. Caveolin-1,amembranesca?oldingproteinfoundinanumberofcellsincludingendothelialandsmoothmusclecells,hasbeen implicatedinPH.Lossofendothelialcaveolin-1isreportedinclinicalandexperimentalformsofPH.Caveolin-1,alsoknown asatumor-suppressorfactor,interactswithanumberoftransducingmoleculesthatresideinorarerecruitedtocaveolae,andit inhibitscellproliferativepathways.Notsurprisingly,therescueofendothelialcaveolin-1hasbeenfoundnotonlytoinhibitthe activationofproliferativepathwaysbutalsotoattenuatePH.Recently,ithasemergedthatduringtheprogressionofPH,enhanced expressionofcaveolin-1occursinsmoothmusclecells,whereitfacilitatescellproliferation,thuscontributingtoworseningofthe disease.Thispapersummarizesthecell-speci?cdualroleofcaveolin-1inPH. 1.Introduction signal transducer and activator of transcription 3 (PY- STAT3) [14, 15], RhoA/Rho kinase [16, 17], and anti- apoptoticmoleculessuchasBcl2andBcl-xL[18,19]have beenreportedinPH.Inaddition,increasedelastaseactivity [20]andincreasedproductionofmatrixmetalloproteinase 2 (MMP2) [21] occur in PH.
