Storm* DepartmentofPharmacology,UniversityofWashington,Seattle,Washington,UnitedStatesofAmerica Abstract Background:ArecentstudyofobesityinSwedishmenfoundthatpolymorphismsinthetype3adenylylcyclase(AC3)are associated with obesity, suggesting the interesting possibility that AC3may play arole in weight control. Therefore, we examinedtheweightofAC3miceoveranextendedperiodoftime. Methodology/PrincipalFindings:WediscoveredthatAC32/2micebecomeobeseastheyage.AdultmaleAC32/2mice areabout40%heavierthanwildtypemalemicewhilefemaleAC32/2are70%heavier.TheadditionalweightofAC32/2 miceisduetoincreasedfatmassandlargeradipocytes.Beforetheonsetofobesity,youngAC32/2miceexhibitreduced physicalactivity,increasedfoodconsumption,andleptininsensitivity.Surprisingly,theobesityofAC32/2miceisnotdueto alossofAC3fromwhiteadiposeandadecreaseinlipolysis. Conclusions/Significance:WeconcludethatmicelackingAC3exhibitobesitythatisapparentlycausedbylowlocomotor activity,hyperphagia,andleptininsensitivity.ThepresenceofAC3inprimaryciliaofneuronsofthehypothalamussuggests thatcAMPsignalsgeneratedbyAC3inthehypothalamusmayplayacriticalroleinregulationofbodyweight.
