RanBPMIsanInhibitorofERKSignaling ElnazAtabakhsh,CarolineSchild-Poulter* RobartsResearchInstituteandDepartmentofBiochemistry,SchulichSchoolofMedicineandDentistry,TheUniversityofWesternOntario,London,Ontario,Canada Abstract Ran-bindingproteinM(RanBPM)isanucleocytoplasmicproteinofyetunknownfunction.Wehavepreviouslyshownthat RanBPMinhibitsexpressionoftheanti-apoptoticfactorBcl-2andpromotesapoptosisinducedbyDNAdamage.Herewe showthattheeffectsofRanBPMonBcl-2expressionoccurthrougharegulationoftheERKsignalingpathway.Transient andstabledown-regulationofRanBPMstimulatedERKphosphorylation,leadingtoBcl-2up-regulation,whilere-expression ofRanBPMreversedtheseeffects.RanBPMwasfoundtoinhibitMEKandERKactivationinducedbyectopicexpressionof activeRasV12.ActivationofERKbyactivec-RafwasalsopreventedbyRanBPM.ExpressionofRanBPMcorrelatedwitha markeddecreaseintheproteinlevelsofectopicallyexpressedactivec-Rafandalsoaffectedtheexpressionofendogenous c-Raf.RanBPMformedacomplexwithbothactivec-Raf,consistingoftheC-terminalkinasedomain,andendogenousc-Raf inmammaliancells.Inaddition,RanBPMwasfoundtodecreasethebindingofHsp90toc-Raf.Finally,weshowthatlossof RanBPM expression confers increased cell proliferation and cell migration properties to HEK293 cells. Altogether, these findingsestablishRanBPMasanovelinhibitoroftheERKpathwaythroughaninteractionwiththec-Rafcomplexanda regulationofc-Rafstability,andprovideevidencethatRanBPMlossofexpressionresultsinconstitutiveactivationofthe ERKpathwayandpromotescellulareventsleadingtocellulartransformationandtumorigenesis.
