Methodology/PrincipalFindings:Anovelpyrosequencingassaywasdevelopedfortherapididentificationandsubtyping of potential human influenza A virus reassortants based on all eight gene segments of the virus. Except for HA and NA genes,oneuniversalsetofprimerswasusedtoamplifyandsubtypeeachofthesixinternalgenes.Withthismethod,all eightgenesegmentsof57laboratoryisolatesand17originalspecimensofseasonalH1N1,H3N2and2009H1N1pandemic viruseswerecorrectlymatchedwiththeircorrespondingsubtypes.Inaddition,thismethodwasshowntobecapableof detecting reassortant viruses by correctly identifying the source of all 8 gene segments from three vaccine production reassortantvirusesandthreeH1N2viruses. Conclusions/Significance:Insummary,thispyrosequencingassayisasensitiveandspecificprocedureforscreeninglarge numbersofvirusesforreassortmenteventsamongstthecommonlycirculatinghumaninfluenzaAviruses,whichismore rapidandcheaperthanusingconventionalsequencingapproaches.
