Methods and Findings: Female goldfish were injected (i.p.) with DA agonists (D1-specific; SKF 38393, or D2-specific; LY 171555)andsacrificedafter5h.SerumLHlevelswerereducedby57%and75%bySKF38393andLY171555,respectively, indicatingthatthetreatmentsproducedphysiologicallyrelevantresponsesinvivo.Bioinformaticstrategiesandaray-finned fishdatabasewereestablishedformicroarrayandiTRAQproteomicanalysisofthehypothalamus,revealingatotalof3088 mRNAsand42proteinsasbeingdifferentiallyregulatedbythetreatments.TwentyoneproteinsandmRNAscorresponding totheseproteinsappearedonbothlists.ManyofthemRNAsandproteinsaffectedbythetreatmentsweregroupedinto theGeneOntologycategorizationsofproteincomplex,signaltransduction,responsetostimulus,andregulationofcellular processes.Therewasa57%and14%directionalagreementbetweenthedifferentially-regulatedmRNAsandproteinsfor SKF38393andLY171555,respectively. Conclusions:Theresultsdemonstratetheapplicabilityofadvancedhigh-throughputgenomicandproteomicanalysesinan amendablewell-studiedteleostmodelspecieswhosegenomehasyettobesequenced.WedemonstratethatDArapidly regulatesmultiplehypothalamicpathwaysandprocessesthatarealsoknowntobeinvolvedinpathologiesofthecentral nervoussystem.
