Results:ExpressionoftheRBCS1genewasanalysedintheallopolyploidcontextofC.arabica,whichoriginates fromanaturalcrossbetweentheC.canephoraandC.eugenioidesspecies.Ourstudyrevealedtheexistenceoftwo homeologousRBCS1genesinC.arabica:onecarriedbytheC.canephorasub-genome(calledCaCc)andtheother carriedbytheC.eugenioidessub-genome(calledCaCe).Usingspecificprimerpairsforeachhomeolog,expression studiesrevealedthatCaCewasexpressedinC.eugenioidesandC.arabicabutwasundetectableinC.canephora. Ontheotherhand,CaCcwasexpressedinC.canephorabutalmostcompletelysilencedinnon-introgressed (pure”)genotypesofC.arabica.However,enhancedCaCcexpressionwasobservedinmostC.arabicacultivars withintrogressedC.canephoragenome.Inaddition,totalRBCS1expressionwashigherforC.arabicacultivarsthat hadrecentlyintrogressedC.canephoragenomethanfor“pure”cultivars.Forbothspecies,waterstressledtoan importantdecreaseintheabundanceofRBCS1transcripts.Thiswasobservedforplantsgrownineither greenhouseorfieldconditionsundersevere
