SpeciesDiscrimination,PopulationStructureand LinkageDisequilibriuminEucalyptuscamaldulensisand EucalyptustereticornisUsingSSRMarkers ShanmugapriyaArumugasundaram1,ModhumitaGhosh1,SivakumarVeerasamy2 ,Yasodha Ramasamy1* 1Division ofPlant Biotechnology, Institute ofForestGenetics andTreeBreeding,Coimbatore, Tamilnadu,India, 2DivisionofGenetics andTreeBreeding, Institute of ForestGeneticsandTreeBreeding,Coimbatore,Tamilnadu,India Abstract EucalyptuscamaldulensisandE.tereticornisarecloselyrelatedspeciescommonlycultivatedforpulpwoodinmanytropical countriesincludingIndia.Understandingthegeneticstructureandlinkagedisequilibrium(LD)existinginthesespeciesis essentialfortheimprovementofindustriallyimportanttraits.Ourgoalwastoevaluatetheuseofsimplesequencerepeat (SSR)lociforspeciesdiscrimination,populationstructureandLDanalysisinthesespecies.Investigationswerecarriedout with the most common alleles in 93 accessions belonging to these two species using 62 SSR markers through cross amplification.Thepolymorphicinformationcontent(PIC)rangedfrom0.44to0.93and0.36to0.93inE.camaldulensisand E.tereticornisrespectively.Acleardelineationbetweenthetwospecieswasevidentbasedontheanalysisofpopulation structureandspecies-specificalleles.SignificantgenotypicLDwasfoundinE.camaldulensis,whereinoutof135significant pairs,17pairsshowedr2$0.1.Similarly,inE.tereticornis,outof136significantpairs,18pairsshowedr2$0.1.Theextentof LD decayed rapidly showing the significance of association analyses in eucalypts with higher resolution markers. The availabilityofwholegenomesequenceforE.grandisandthesyntenyandco-linearityinthegenomeofeucalypts,willallow genome-widegenotypingusingmicrosatellitesorsinglenucleotidepolymorphims.
