FAP与E―cadherinN―cadherin在大肠癌中表达及相关性研究 　　[摘要] 目的 探讨FAP与E-cadherin＼N-cadherin的表达与大肠癌发生发展的关系及其相关性。方法 采用免疫组化EN法检测150例大肠癌标本中FAP与E-cadherin ＼N-cadherin的表达。结果 FAP表达与肿瘤分期及淋巴结转移呈正相关，差异有统计学意义（P0.05）。E-cadherin的阴性表达率及N-cadherin阳性表达率与大肠癌的肿瘤分级、分期及淋巴结转移呈正相关，差异有统计学意义（P0.05）。E-cadherin ＼N-cadherin阳性表达呈负相关，差异有统计学意义（r = -0.745，P0.05 ）。 结论 FAP高表达、E-cadherin的低表达或不表达和N-cadherin的反常表达与大肠癌的浸润＼恶化有密切关系。 　　[关键词] 大肠癌；FAP；E-cadherin；N-cadherin 　　[中图分类号] R4 [文献标识码] A [文章编号] 1674-0742（2015）06（c）-0010-02 　　[Abstract] Objective To explore the correlation between expressions of FAP and E-cadherin＼N-cadherin as well as their relationship with the carcinogenesis in colorectal carcinoma. Methods The expression of FAP and E-cadherin＼N-cadherin was detected by immunohistochemical EN technique in 150 cases of colorectal carcinoma. Results The expression of FAP were positively associated with tumor staging and lymph node metastasis（P0.05. Negative expression rate of E-cadherin and positive expression rate of N-cadherin were positively associated with tumor staging， tumor grading and lymph node metastasis（P0.05）.The expression of E-cadherin was negatively correlated with N-cadherin expression （r = -0 .745，P 0.05）. Conclusion Over-expression of FAP， negative or weak expression of E-cadherin， and abnormal expression of N-cadherin were have significantly correlated with invasion/aggravation of colorectal carcinoma. 　　[Key words] Colorectal carcinoma；FAP；E-cadherin；N-cadherin 　　FAP是分子量97KDa的细胞表面糖蛋白，特异性的表达在癌相关纤维母细胞中。通过抗体免疫疗法靶向灭活FAP活性达到抑制肿瘤侵袭转移的目的。E-cadherin 和N-cadherin是钙依赖黏附素家族中的成员，分布于上皮细胞相连接的细胞膜上，其表达减弱或无表达可导致癌细胞脱落并发生浸润转移。该研究收集黑龙江省医院病理科2012年1―12月大肠癌手术病例150例，探讨FAP与E-cadherin 和N-cadherin 表达与大肠癌浸润＼ 恶化的关系及它们在大肠癌中表达的相关性，进一步明确其临床意义。 　　1 资料与方法 　　1.1 标本来源 　　随机收集黑龙江省医院病理科2012年1―12月的大肠癌手术病例150例，男性90例，女性60例，年龄26～84岁，平均年龄56，标本经10%甲醛固定，石蜡包埋，4 um切片，HE染色。FAP购自美国Abcam公司，鼠抗人E-cadherin 及N-cadherin单克隆抗体购自北京中山生物技术开发中心。 　　1.2 实验方法 　　免疫组化染色采用EnVision法。 　　1.3 结果判定 　　FAP表达于肿瘤基质细胞的细胞质。E-cadherin ＼ N-cadherin 表达于肿瘤细胞的细胞膜或质。
