AbstractAcomprehensiveanalyticalmethodbasedonsolid—phaseextraction(SPE)andultra
performanceliquidchromatography—electrospraytandemmassspectrometry(UPLC—MS/MS)has
beendevelopedforthesimultaneousdeterminationofsevenglucocorticoidsinwastwatersample.
Severalsinglefactorexperimentswereappliedtoanalyzetheeffectofthesolventandvolumeusedfor
elution,pHadjustmentofthewatersample,andthewashsolutiononSPE.Basedonthesingle
factortests,theL9(3)orthogonaltestwasusedtofurtheroptimizeextractionparameters.The
primaryandsecondaryfactorsweredistinguishedbyvisualanalysisandanalysisofvariance.The
optimumextractionprocesswasasfollows:elutingsolventwasethylacetate,elutionvolumewas
10mI,thepHvaluewas5.0andwashingsolventwas2O(v/v)methanolinHPLCgradewater.
Thecorrelationcoefficient(r.)ofthecalibrationcurvewasover0.9976,inlinearrangeof1—
100gg/L.Thelimitsofdetection(LOD)werebetween1.56and10.59ng/L.Therecoveriesatthe
spikedlevelof20—100ng/Lwere72.48%to101.42,withtherelativestandarddeviation(RSD)less
than10.9.
