维普资讯 JReprodM ed2007Vo1．16No．S1 TheexpressionofTGF—pmRNAanditsbiologicalfunctioninrat osteoblastswith exposuretoraloxifene GuoLing-cen(郭凌岑)，LiuJia-yin(刘嘉茵)“，ZhangKe-qin(张克勤)， ZhangHong-mei(张红梅)，GongJian(龚健) 0bjective：Toinvestigatetheeffectsofraloxifeneandestradiolontheproliferation，differentiationandthe expressionoftransforminggrowthfacto叫3(TGF-p)ofosteoblastsinvitro． Methods：Differentdosesofraloxifeneand estradiolwereadded into themedium ofthesecond generation osteoblastsobtained from theskullofnewborn SD rats． Thefollowing parameters including cellproliferation， activityofalkalinephosphatase(ALP)，thelevelsoftypeIcollagen(Col_I)mRNAandTGF-~31mRNA indifferent groupsweremeasuredandanalyzed． Results：Raloxifeneand17一~estradiol(17一BEz)showednosignificanteffectonstimulatingtheproliferationof osteoblasts(P 0．05vsthecontro1)．However，raloxifenecouldsignificantlyimproveALP activityand Col—I mRNA expressioninhighconsistencygroup(PO．O1)indose-dependentmanner．Raloxifenegroupin10 mol／L increasedtheexpressionofTGF-~ImRNA (vsthecontrol，P 0．05)．EstradiolsignificantlyincreasedALP activity，Col—ImRNA expressionandTGF-[31mRNA expression(P0．05orP0．O1vsthecontro1)． Conclusions：Both ofraloxifeneand estradiolcouldstimulatedifferentiation ofosteoblastsand expression of bonematrix，butshowednoeffectonproliferationofculturedosteoblasts．TheexpressionsofTGF-13】mRNA were different，whichmightimplytheirdifferentmechanismsbymeansofestrogenreceptor． Keywords：Osteoblast；Raloxifene；Estradiol；TGF-[3 CLC nnmber：R711 Documentcode：A ArtieleID：1004—3845(2007)S1—0084—05 postmenop
