Medicine Enzymatic Assay of D-serine Background D-serine functions as a co-agonist of the N-methylic-D-aspartic acid (NMDA) receptor in the mammalian brain. In the serum of Alzheimers disease patients, the ratio of D-serine to total serine content is significantly low. D-serine’s relation to neurological disorders has been collecting much attention these days. Despite promising D-serine applications in the future, there are currently no simple D-serine quantification method. Technology Researchers at Nagoya University found a novel enzyme, D-serine dehydratase, which can be used to measure the D-serine. The quantification methods consists of (1) reacting the sample with D-serine dehydratase to decompose D-serine into pyruvic acid or ammonia, (2) measuring pyruvic acid by using lactate dehydrogenase; or measuring ammonia by using pyruvate oxidase, and (3) quantifying D-serine via the data obtained in (2) Advantages Simple: No complicated pre-treatment is needed Precise: Almost free from L-amino acid noise Easy: No high proficiency required to analyze the data Efficient: Large quantity of sample can be processed in bulk Safe: Disposable; no infection risk Cheap: Expensive instruments used in High-performance liquid chromatography (HPLC), capillary electrophoresis, or gas chromatography are not needed Applications D-Serine Assay, Diagnostic Kit for Schizophrenia and Alzheimer’s Disease Patent: PCT/JP/2007/070193, WO2008/047802, US2010/0190196,
