广 东 化 工 2006 年 第 1 期 ·42 · www . gdchem. com 第33 卷 总第153 期 插入式超声对酵母细胞膜 通透性影响的研究 姚咏嫦 （广东石油化工设计院，广东广州510 130 ） ［摘 要］本论文选择生产1，6 - 二磷酸果糖（FDP ）的酵母细胞为研究对象，采用紫外分光光度计测定超声 处理后酵母细胞内核酸和蛋白质透出细胞膜的情况，使用次甲基蓝对超声处理后的细胞进行染色，用血球计数， 测试经超声处理后细胞的生存能力，及通过测定胞外FDP 的浓度变化，反映细胞膜通透性的变化。结果表明， 插入式超声对细胞的通透性及细胞的存活率都有很大影响，而脉冲超声的间隙时间对核酸、蛋白质及细胞活性 影响不是很大，但对FDP 的渗出率有较大影响。 ［关键词］超声；细胞膜；通透性 Effect of Probe Ultrasound on Yeast Cell Membrane Permeability Yao Yongchang （Guangdong PetrochemicaI Engineering Design Institute ，Guangzhou 510 130 ，China ） Abstract ：Yeast ceII ，which can produce Fructose-1，6-diphosphate （FDP ），was chosen as object. Inner-ceII nu- cIeic acid and protein penetrating membrane after the disposaI of uItrasound was measured by uItravioIet radiation spec- trometer to determine the extent of effect on ceII membrane penetrate. CeIIs treated by uItrasound were dyed by MethyI- ene BIue SoIution and counted by haemacytometer to examine the viabiIity of the ceIIs. And the variety of membrane per- meabiIity was determined by testing the variety of FDP content in the broth. The resuIts showed that，probe uItrasound had great effect on ceII permeabiIity and IivabiIity. The intermittent time of puIsed uItrasound has IittIe infIuence on the transport of nucIeic acid，protein and on ceII activity. But it had great effect on the permeabiIity of FDP.
