Methods We transfected the recombinant plasmids pEGFP-C1-T8-Sense and pEGFP-C1-T8- AntiSense into HL60 with liposomes, and studied the variational characteristics of MMP2, MMP14 and TGF- 1 expression on mRNA and protein level with RT-PCR and Western blot. The invasive potential of the transfected cells was examined with the flow transwell plate. Results The expression of 3GnT8 increased in HL60 transfected with sense recombinant plasmid while the expression of MMP2 and MMP14 also upgraded. 3GnT8 expressed less in HL60 transfected with antisense recombinant plasmid while MMP2 and MMP14 also expressed weaker. 3GnT8 has negative correlation with TGF- 1.Cell invasion ability is higher than that of the control group (P 0.05).
