[Abstract] Objective: To observe the post-severe traumatic brain injury in rats COX 2 expression and selective COX 2 inhibitor NS 398 after severe traumatic brain injury in rats learning and memory function. METHODS: Marmarou s method Establishment of rat model of severe closed traumatic brain injury, 360 adult Wistar rats were randomly divided into the brain injury group, NS 398 treatment group, sham operation group, each group was divided into injury 1, 3, 6, 12, 24, 48, 72, 96, 168 and 336 h 10 hour-time-phase group, and the other to take 120 as a normal control group. NS 398 through the injury after the treatment group given intraperitoneal injection of NS 398 40 mg / (kg * d ), until killed at different time points; brain injury group, sham operation group and the normal control group at the same time giving the same amount of saline intraperitoneally. in each group were killed at the corresponding point in time derived method of application of Western Blot and Immunohistochemistry Detection of COX 2 protein changes. and then take 24 rats were randomly divided into the brain injury group, NS 398 treatment group, sham operation group and normal control group, for water maze test. Results: NS 398 enable the rat brain COX 2 protein expression within the peak of significantly down-regulated, so that the water maze test was significantly shorter incubation period. Conclusion: COX 2 and post-traumatic brain injury-induced secondary brain injury are closely related, NS 398 can be significantly improved in rats with severe head post-traumatic learning and memory impairment.
