Immunoprecipitation identified monomeric and oligomeric pUL77 in HCMV infected cells. Immunostaining of purifiedvirionsandsubviralfractionsshowedthatpUL77isastructuralproteinassociatedwithcapsids.Insilicoanalysis revealedthepresenceofacoiled-coilmotif(CCM)attheN-terminusofpUL77.Chemicalcross-linkingofeitherwild-type pUL77orCCMdeletionmutant(pUL77DCCM)implicatedthatCCMiscriticalforoligomerizationofpUL77.Furthermore,co- immunoprecipitationsofinfectedandtransfectedcellsdemonstratedthatpUL77interactswiththecapsid-associatedDNA packagingmotorcomponents,pUL56andpUL104,aswellasthemajorcapsidprotein.TheabilityofpUL77tobinddsDNA wasshownbyaninvitroassay.BindingtocertainDNAwasfurtherconfirmedbyanassayusingbiotinylated36-,250-,500-, 1000-mericdsDNAand966-mericHCMV-specificdsDNAdesignedforthisstudy.Thebindingefficiency(BE)wasdetermined byimageprocessingprogramdefiningvaluesabove1.0aspositive.WhiletheBEofthepUL56bindingtothe36-merbio- pac1containingapackagingsignalwas10.060.63,theoneforpUL77wasonly0.260.03.Incontrasttothisobservationthe BEofpUL77bindingtobio-500bporbio-1000bpwas2.260.41and4.960.71,respectively.ByusingpUL77DCCMitwas demonstratedthatthisproteincouldnotbindtodsDNA.ThesedataindicatedthatpUL77(i)couldformhomodimers,(ii) CCMofpUL77iscrucialforoligomerizationand(iii)couldbindtodsDNAinasequenceindependentmanner.
