Using a mathematical model, we investigate how different synaptic transmission strategies, defined by the numberofvirusespassedpersynapse,influencethebasicreproductiveratioofthevirus,R0,andvirusload.Inthemost basicscenario,themodelsuggeststhatR0ismaximizedifasinglevirusparticleistransferredpersynapse.R0decreasesand theinfectioneventuallycannotbemaintainedforlargernumbersoftransferredviruses,becausemultipleinfectionofthe samecellwastesvirusesthatcouldotherwiseenteruninfectedcells.ToexplaintherelativelylargenumberofHIVcopies transferred per synapse, we consider additional biological assumptions under which an intermediate number of viruses transferredpersynapsecouldmaximizeR0.Theseincludeanincreasedburstsizeinmultiplyinfectedcells,thesaturationof anti-viralfactorsuponinfectionofcells,andratelimitingstepsduringtheprocessofsynapseformation.
