Methods:Toevaluatethediagnosticvalueofthisproteininhepatictumorswehaveinvestigatedthepresenceof EZH2byimmunohistochemistryinhepatocellularcarcinomasandothercommonhepatictumors. EZH2expressionwasexaminedin44hepatocellularcarcinomas,23cholangiocarcinomas,31hepatoblastomas,16 otherchildhoodtumortypes(rhabdomyosarcoma,neuroblastoma,Wilms’tumorandrhabdoidtumor),17 metastaticlivertumors24hepatocellularadenomas,15highgradedysplasticnodules,3biliarycystadenomas,3 biliaryhamartomasand3Caroli’ sdiseases. Results:MostofthemalignantlivertumorswerepositiveforEZH2,butneitheroftheadenomas,cirrhotic/ dysplasticnodules,reactiveandhamartomatousbiliaryductulesstainedpositively. Conclusions:OurimmunostainingsconfirmthatEZH2isasensitivemarkerofhepatocellularcarcinoma,butits specificityisverylow,sincealmostalltheinvestigatedmalignantlivertumorswerepositiveregardlessoftheir histogenesis.BasedontheseresultsEZH2isasensitivemarkerofmalignancyinhepatictumors.Inroutinesurgical pathologyEZH2couldbemosthelpfultodiagnosecholangiocarcinomas,becauseasfarasweknowthisisthefirst markertodistinguishtransformedandreactivebiliarystructures.AlthoughhepatoblastomasalsoexpressEZH2,the diagnosticsignificanceofthisobservationseemstobequitelimitedwhereas,thestructurallysimilar,otherblastic childhoodtumorsarealsopositive.
