163 16(3) :270- 274 20019 VIROLOGICA SINICA September 2001 周鹏程, 聂玉春, 曹圻, 陈建国, 丁明孝 (, 100871) Study on the Inhibition of Specific 01igonucleotides on Classical Swine fever Virus in vitro Z OU Pengcheng,NIE Yuchun,CAO Qi,C EN Jianguo,DING Mingxiao ( Colleg e of Lfi e Science, P eking University , Beij ing 100871 , China) Abstract:The effect of oligodeoxynucleotides(ODN) on replication of CSFV(Shimen strain) was investigated in this paper.Phosphorthioate 01igodeoxynucleotides ( PSODN) targeted to the region of the 3 ,5noncoding region(NCR) as well as PSODN targeted to NS3 gene region can markedly inhibited the replication of CSFV in PK15 cells.Antiviral activity of PSODN was evaluated by fluorescent plaque formation assay.The results presented here indicated that ODN P5 complementary to 5 terminal conserved sequence and it s sense ODN P5S, as well as P8 complementartoNS3 region exhibited strongly antiviral activity,whereas theantiviralactivi ty of P3 and it s enese PSODN was much lower.At the sametime,antiviral activity of senseODN is slightly higher than corresponding antisenseODN.The inhibition of ODN is in a doseand sequencedependantmanner. Inhibition of CSFV s replication in PK15 cells can be signfiicantly increased in the presense of lipofectin by either sense or antisenseODN.These data suggested both 5NCR and 3NCR may play important roles in CSFV replication.
