河川沙塘鳢雌雄亲鱼肌肉中脂肪酸含量的比较研究 Abstract [Objective] The research aimed to compare the content of fatty acid in the muscles of female and male Odontobutis potamophila. [Method] Taking female and male ontobutis potamophila in different reproductive stages（III，IV and V stages） as the research objects， the changes of fatty acid composition in the muscles were analyzed by using chloroform-methanol extraction method and gas chromatography.[Result] 27 kinds of fatty acids were detected in the muscles of male and female O. potamophila and there was rich unsaturated fatty acids in the muscles. The content of n-3 fatty acids remained unchanged in the muscles of female O. potamophila. The content of n-6 fatty acids reached the maximum at III stage and the minimum at V stage. n-6 fatty acids were mainly used the development of ovaries. In the muscles of male O. potamophila， the content of n-6 fatty acids reached the maximum at IV stage， which stored energy for the development of testis. The content of n-3 fatty acids in the muscles of female . potamophila was higher than that of n-6 fatty acids. [Conclusion] The research provided theoretical guide for the artificial culture of O. potamophila. Key words Odontobutis potamophila；Fatty acid content；Male and female fish；Comparison 河川沙塘鳢（Odontobutis potamophila），隶属鲈形目（Perciformes）沙塘鳢科（Odontobutidae），分布在我国长江中下游及其支流、钱塘江水系、闽江水系等，是我国江苏太湖地区最为常见的底栖肉食性经济鱼类，深受沿海地区人们的喜爱[1-2]。大力发展河川沙塘鳢的人工增养殖具有重要的经济意义。目前，有关河川沙塘鳢的研究主要集中在早期发育[3-6]，其中脂类含量及脂肪酸组成的研究主要集中在胚胎和仔鱼时期，而对亲鱼的脂类含量及脂肪酸组成的研究较少。笔者以不同生殖时期的河川沙塘鳢雌、雄鱼亲鱼为研究对象，分析其肌肉的脂肪酸组成的变化，旨为开展河川沙塘鳢的人工增养殖提供理论指导。 1 材料与方法 1.1 试验材料 试验材料为河川沙塘鳢雌、雄亲鱼，2012 年 4 月自上海市铜川路水产品集贸市场购买。根据性腺发育周期，分别在III、IV和V期各取样 8 尾，取其背部肌肉保存于-80 ℃低温冰箱中，待测。 1.2 试验方法 1.2.1 脂肪酸组分的测定。参照国家标准GB/T*****-2008，采用水解提取-气相色谱法测定。 1.2.1.1 脂肪酸的提取。准确称取均匀捣碎的样品1 g，置于玻璃离心管内，依次加氯仿 20 ml、甲醇10 ml、水 2 ～ 3 ml，盖上盖子，超声 10 min 后再振荡 2 h。静置 22 h 后，3 500 r/min 离心 10 min，分层后取下层氯仿层，过滤纸 （滤纸上放 20 g 烘干过的无水硫酸钠） ，滤液置于干燥的烧瓶中，在真空干燥箱内烘干。 1.2.1.2 脂肪酸的甲酯化。在盛有脂
