Phosphatebuffer(pH7)-Takepotassiumdihydrogenphosphate2.713gandmakeupto1000mL

withwater.Adjustwith1mol/LsodiumhydroxidesolutiontoobtainpH7.

Standarddihydrostreptomycinsulphate–1.000mgofthisproducthasatleast0.720mgof

dihydrostreptomycin

Standardstreptomycinsulphate–1.000mgofthisproducthasatleast0.718mgofstreptomycin

4.Procedure

1)Extraction

Take20.0gofsampleandadd100ml1%metaphosphoricacidand50mLdichloromethane.After

homogenizing,separateitcentrifugallyunder3,000rpm.Collecttheaqueouslayer.Add50mL1%

metaphosphoricacidintothedichloromethanelayerandresidue.Mixandrepeatthecentrifugal

separation.Collecttheaqueouslayerandcombinewiththepreviousaqueouslayer.Makeupto

200mLwith1%metaphosphoricacid.

2)Clean-up

(i)OctadecylsilanizedSilicaGelColumnChromatography

(Sep-PakPlusShortC18,360mg,P/N:WAT020515)

Add5mLofmethanoland5mLof1%metaphosphoricacidintotheoctadecylsilanizedsilicagel

mini-column(360mg).Discardtheeffluent.Afteradding20mLofthesampleobtainedfrom

extractionstep1),add5mL1%metaphosphoricacid.Collectalleffluenta
