D ep a rtm en t of Op h tha lm ology, Un ion H osp ita l, Tongj i M ed ica l Colleg e, H uazhong Un ivers ity of S cience and Technology, W uhan 430022, Ch ina 　　A b stract　O bjective　The aim of th is p ap er was to study the inh ib itory m echan ism of CTLA4dendritic cells ( CTLA4 DC) in corneal tran sp lan tation. 　M ethods　The p la sm id PG/CTLA4 Ig wa s tran sferred in to the dendritic cells of F344 rat m ediated by L ipofectam ineTM 2000. The change of the exp re ssion of indo lam ine 2 , 3 dioxgena se ( IDO ) mRNA in dendritic cells wa s m ea sured by u sing sem iquan tu tative reverse tran scrip tion po lym erase chain reaction. The p roliferative re spon se s of T cells in L ew is rat by the effect of the tran sfected dendritic cells were determ ined u sing CCK8. The dendritic cells (48 hours after tran sfer) were divided in to two group s, and m ixed lymp hocyte reaction (MLR ) wa s p erform ed between dendritic cells and T cell. L tryp top han wa s p ut into the exp erim ent group , and T cell apop to sis was determ ined by the flow cytom etry assay in A nnexinV / P I. Corneal tran sp lan tation was p erform ed from F344 rats to L ew is rat, and the dendritic cells of above two group s were injected in to the L ew is rat after the op eration.
