山西医科大学硕士学位论文 CerbB-2 基因在胃腺癌中的表达与扩增及其意义 摘 要 目的：研究原癌基因CerbB-2在胃腺癌发生、发展过程中的意义。 方法：应用免疫组织化学法（IHC）和荧光原位杂交技术（FISH）对60例胃腺癌组织标本、 60例正常切缘粘膜组织标本和30例正常胃粘膜组织标本中CerbB-2基因的表达与扩增情况进 行检测。 结果：CerbB-2基因在胃腺癌组织、正常切缘组织和正常胃粘膜组织中的表达阳性率分别 为31.7%、8.33%、3.33%；扩增阳性率分别为28.3%、5%、0%；胃腺癌组织中CerbB-2基因 的表达、扩增阳性率与正常切缘组织、正常胃粘膜组织相比较，存在显著性差异；在胃腺 癌组织中，CerbB-2基因的表达和扩增与患者的年龄、性别、肿瘤大小、分化程度均无相 关性（P＞0.05），而与肿瘤的浸润程度、淋巴结转移、远处转移、临床分期相关（P＜0.05）。 结论：CerbB-2基因与胃腺癌的发生、发展有明显的关联，可以将其作为判断胃癌的生物性 行为的重要参考指标。 关键词：胃腺癌；CerbB-2；荧光原位杂交 I 山西医科大学硕士学位论文 The expression and amplification of CerbB-2 oncogene in the gastric adenocarcinoma and its significance Abstract Objective:To investigate the significance of CerbB-2 oncogene in the occurrence and development of gastric adenocarcinoma. Methods:By using immunohistochemistry and fluorescence in situ hybridization, 60 cases of gastric adenocarcinoma tissue, 60 cases of the normal margin of mucosal tissue and 30 cases of normal gastric mucosa tissue were detected the expression and amplification of CerbB-2. Results:The positive rate of CerbB-2 expression in gastric adenocarcinoma tissues,normal margin of mucosal tissues and normal gastric mucosa tissues were 31.7%,8.33% and 3.33%; The positive rate of CerbB-2 amplification of these were 28.3%, 5% and 0%. There were significantly differences between gastric adenocarcinoma tissues and the others in the expression and amplification of CerbB-2.And the expression and amplification of CerbB-2 in gastric adenocarcinoma tissues had no correlation with age,gender,tumor
