College of Life Sciences, Agricult ural University of H ebei, Baoding 0 1001, China) Abstract: Genomic in s i tu hybridization ( GISH) may be improved if suitable blocking DNA is used. Chinese cabbage genomic DNA w as cut by boiling and ultrasonic method to study the preparation of blocking DNA. T he results show ed that the efficiency of boiling method w as higher than ultrasonic cutting method, and boiling method w as more easily operated. Most DNA length w as 200- 500 bp w hen genomic DNA w as boiled for 0 min, thus DNA w as suit- able for blocking DNA in GISH. T hese findings laid a foundation for further study on GISH.
