ChineseMedicalJournal2014：127(i3) 2463 — — Originalarticle Heparininhibitsburn-inducedspleencellapoptosisbysuppressing i0nterlleullo-n-1eXDressl●on ZhaoSongfeng，ZhangXiao，ZhangXiaojian，ShiXiuqin，YuZujiangandKanQuancheng Keywords：burn,spleen，apoptosis，‘heparin，。interleukin—J『 BackgroundEpidermalburninjurymaytriggersignificantapoptosisofthespleencells，whichmightbecausedbyaburn． induced systemicinflammatory reaction．Heparinhasbeenshowntopossessanti．inflammatoryproperties ． 1nterleukin1 (1L_1)iscentrallyimportantamongpro-inflammatorycytokines．Wehypothesizedthatheparinmightinhibitburn．induced apoptosisinthespleenviasuppressionoftheIL．1pathway． Methods BurninjurywasperformedonIL一1R+／+(IL一1receptorwild．typemouse)andIL．1R-／一(IL．1receptorknock． outmouse)mice，andtheywerethentreatedwithheparin，salineorlL-1receptorantagonistlLRa．Apoptosis，lL．1aand IL-113expressionwereassessedinthespleensandserum．SurvivaIcurveanalysiswasfurtherappliedtoelucidatethe mechanism ofheparin’sprotectiveproperties． Results Burninducedsignificantapoptosis(sham：3．6％±2．1％vs．burn：28．8％±5．9％：P0．001)andremarkable expressionoIL-1aandlL-131inthemousespleensandserum，Heparinreducedtheburn．inducedapoptosisinthespleens (heparintreated：8_6％±3．4％，P0．005)，whichcouldbeblockedbylL．1Ra．HeparinmarkedlydecreasedbothIL．1aand lL-131expressioninthespleensandserumofburnedmice．fL．1R-／-micedemonstratedconsiderablyIessapoptosisinthe spleensandhadahighersurvivaIrateafterburns．Heparindidnotsignificantlydecreaseapoptosisinthespleenandthe mortalityrateinlL一1R4-miceafterburns． Conclusion Heparininhibitsburn—inducedapoptosisofthespleencellsbysuppressinglL．1expressioninmice．
