内蒙古科技大学本科生 毕业论文 论文题目：NF 学生姓名：谢强强 学 号：1168149122 专 业：生物技术 班 级：生技2011 指导老师：李珺副教授 NF1-A Minigene构建与鉴定 摘要 人类I型神经纤维瘤病(Neurofibromatosis type I，NF1)是最常见常染色体的显性遗传性肿瘤疾病中的一种，其主要的发病机制为NF1基因的突变缺失。可变剪接剪接因子导致真核基因呈现复杂程度从而显示蛋白质拥有多样性。本论文主要论述了可变剪接的机理，NF1基因的结构与功能，以及NF1前体mRNA的编辑。本实验提取了人体外周血基因组DNA，设计了两对引物，利用PCR方法扩增NF1基因并导入pcDNA3.1质粒载体从而构建了NF1-A minigene基因，为后续NF1的mRNA前体可变剪接以及调控NF1剪接因子的研究奠定了基础。 关键词：可变剪接；NF1基因；载体 Construction and Identification of NF1-A Minigene Abstract Neurofibromatosis type 1 (NF1) is one of the most common heritable autosomal dominantdisorders.Alternative splicing modulates the function of neurofibromin, the NF1 gene product, by inserting the in-frameexon 23a into the region of NF1 mRNA that encodes the GTPase-activating protein-related domain. This paper mainly discusses the mechanism of alternative splicing, the structure and function of NF1 gene, which introduces the variable splicing of mRNA precursor NF1 and the splicing factor of NF1 variant splicing in the regulation of the variable splicing.
