This is generally set between 0.5-3.2 kV in ESI positive and 2.6 kV in ESI negative, but can be altered according to sample type. For infusion or low flow applications best sensitivity will be achieved with a relatively high value in ESI positive e.g. 3.0 kV, for standard flow UPLC a value of 0.5 kV is typically best for maximum sensitivity. Sample Cone - The voltage applied to the sample cone. This voltage is sample dependent for sensitivity and does not have any effect on the resolution. The sample cone voltage normally optimizes between 25 and 100V (when maximizing the intensity of the [M+H]+ ion). If it is set too high in-source fragmentation can occur. Extraction Cone – Ions flowing through the sample cone are pulled through the extractor opening into the ion guide. The ions then enter into the analyser region of the mass spectrometer. The extraction cone setting is also optimized for the intensity of the [M+H]+ ion. Typically set between 1 and 5V. Source (°C) - The temperature setting for the source block. Heating the source block discourages solvent clustering in the reduced vacuum region of the source. The temperature is flow rate and sample dependent. T
