间充质干细胞移植后在溃疡性结肠炎大鼠体内的迁移 段 征，田静，汪维伟 （重庆医科大学组织胚胎学教研室，干细胞与组织工程研究室，重庆 400016） 【摘要】 目的 观察大鼠骨髓间充质干细胞（bone marrow mesenchymal stem cells，BMSCs）静脉移植后在体内的迁移。方法 SD大鼠分为A组（免疫结合 TNBS/ethanol诱导溃疡性结肠炎模型）和B组（正常大鼠），均尾静脉注射1mL荧光标记的BMSCs。移植后第3、7、14天各处死5只，取材做恒冷切片，荧光显微镜下观察BMSCs在骨髓、肺、肝、脾以及消化道内分布，测定荧光的积分光密度（IOD）值。结果 BMSCs移植后多分布于血管丰富器官，7d时肺内荧光IOD值最高，14d时骨髓内IOD值最高，但无统计学意义。14d时，A组结肠的IOD值高于同组其余消化道各段以及B组结肠（P0.05）。结论 静脉移植的BMSCs早期主要分布在血供丰富的器官中，迁移至受损结肠的BMSCs高于正常结肠。 【关键词】骨髓间充质干细胞；移植； 溃疡性结肠炎，大鼠 【中国图书法分类法分类号】 R574.1；R-332 【文献标识码】A Endosomatic transference of transplanted bone marrow mesenchymal stem cells in ulcerative colitis rat DUAN zheng, TIAN jing, WANG wei-wei (Department of Histology and Embryology, Laboratory of stem cells and tissue engineering, Chongqing Medical University, Chongqing 400016 ) 【Abstract】 Objective To investigate the transference of transplanted bone marrow mesenchymal stem cells (BMSCs) in ulcerative colitis rat. Methods SD rats were randomly distributed into groups of Colitis induced by immune- combined TNBS/ethanol and groups of control. All rats received caudal vein injection with 1 mL of fluorescence marked BMSCs, then 5 rats were sacrificed at day 3, 7 and 14 in each group. Ccryostat sections of bone marrow, lung, liver, spleen and gut were made to observe the distribution of BMSCs in these organs, and the fluorimetric integral optical density（IOD）were analyzed. Results BMSCs mainly distributed in organs with rich vessel. The highest IOD level of BMSCs in lung was on the 7 day, and was on the 14 day in bone marrow.
