人参皂甙Rh2 致MCF - 7/ ADM凋亡的实验研究 张　晖1 ,2 ,孔　棣2 ,吴咸中2 ,张会来3 ,王华庆3 11 天津医科大学博士研究生(天津300070) 21 天津市中西医结合急腹症研究所 31 天津医科大学肿瘤医院 摘要　目的:本实验旨在研究Rh2 对肿瘤细胞生长的抑制作用及其机理,以进一步阐述Rh2 在抗肿瘤方面的作用。 方法:Rh2 作用于MCF - 7 及MCF - 7/ ADM细胞,应用MTT 法确定各组细胞增殖抑制率。流式细胞仪分析Rh2 导致MCF - 7 和MCF - 7/ ADM细胞坏死及凋亡的作用,及其对细胞周期的影响,并检测Rh2 对MCF - 7 和MCF - 7/ ADM细胞内Caspase 3 活 性的影响。结果:Rh2 可以显著抑制MCF - 7 和MCF - 7/ ADM的增殖( P 0105) ,并有时间和剂量依赖关系,对MCF - 7 抑制 作用更强。Rh2 可以导致MCF - 7 坏死,通过Caspase 途径诱导MCF - 7/ ADM凋亡( P 0105) ,对两种细胞周期的影响也有所不 同。结论:人参皂甙Rh2 通过不同机制抑制了MCF - 7 和MCF - 7/ ADM细胞的增殖,Rh2 促进耐药细胞系MCF - 7/ ADM凋亡 的研究结果显示其在抗肿瘤作用方面的独特性。 关键词:人参皂甙Rh2 ;MCF - 7 细胞系;增殖抑制;凋亡 中图分类号:R 28515 　　　文献标识码: 　A 　　　文章编号:1007 - 6948 (2007) 03 - 0261 - 04 Apoptotic Effects of Ginsenoside Rh2 on Drug Resistance Human Mammary Adenocarcinoma MCF27/ ADM Cells 　Zhang Hui ,Kong Di , Wu Xianzhong , et al . 　Tianjin Medical University , Tianjin (300070) , China Abstract :Objective 　To research on the mechanism of inhibitory effect of ginsenoside Rh2 on human mammary adenocarcinoma MCF27 and the drug resistance cell line MCF27/ ADM in vitro 　Methods 　In MTT tests , Rh2 were added to MCF27 and MCF27/ ADM, inhibition ratios were determined. In flow cytometric analysis , necrotic and apoptotic effect of Rh2 on MCF27 and MCF27/ ADMwere determined. Effects of Rh2 on cell cycle were determined too. The activity of caspase 3 in MCF27 and MCF27/ ADM cell lines was also detected. 　Results 　Ginsenoside Rh2 could inhibit the pro2 liferation of MCF27 and MCF27/ ADM cells with time and dose2dependent ( P 0. 05) ,Rh2 had stronger effect on MCF27 cells. Rh2 could cause necrosis on MCF27 , and induce apoptosis on MCF27/ ADM with Caspase pathway. The effects of Rh2 on Cell cycles on MCF27 and MCF27/ ADMwere different too. 　Conclusion 　Ginsenoside Rh2 can inhibit the pro2 liferation of MCF27 and MCF27/ ADM cells with different mechanism.
