再生障碍性贫血血管内皮生长因子表达及其意义的研究 nbsp;nbsp;nbsp;nbsp;nbsp; 作者：季健玲，刘红，孙超，姜胜华，丁润生 【摘要】nbsp; 本研究目的是探讨再生障碍性贫血骨髓血管内皮生长因子(VEGF)的表达情况，采用逆转录聚合酶链反应（RT-PCR）法半定量检测7例再生障碍性贫血患者和12例正常人骨髓单个核细胞VEGF基因的表达；用免疫组织化学法检测20例再生障碍性贫血患者和20例正常人骨髓组织中VEGF的表达。结果表明，7例再生障碍性贫血患者骨髓标本有2例表达VEGF 基因，表达率为28.57％，而12份正常人骨髓标本有10例表达VEGF基因，表达率为83.33％；再生障碍性贫血患者VEGF基因表达率和表达强度都显著低于正常人，两者差异具有统计学意义（Plt;0.05）；免疫组织化学检测发现，再生障碍性贫血患者骨髓活检组织无1例表达VEGF，而正常人表达率为25％（5/20），再生障碍性贫血患者骨髓活检组织VEGF蛋白表达阳性率低于正常人（Plt;0.05）。结论： 再生障碍性贫血患者骨髓VEGF在基因转录水平和蛋白质表达水平都有显著降低，这可能与再生障碍性贫血患者骨髓血管生成减少和造血减少有一定关系。 【关键词】nbsp; 再生障碍性贫血 　Expression of Vascular Endothelial Growth Factor in Patients with Aplastic Anemia and Its Significance 　Abstractnbsp;nbsp;nbsp; This study was aimed to investigatenbsp; the expression of vascular endothelial growth factor （VEGF） in patients with aplasticnbsp; anemia.Thenbsp; gene expressions of VEGF in mononuclear cells of bone marrownbsp; from 7 cases of aplastic anemia and 12 normal controlsnbsp; were detected by semi-quantitative reverse transcriptase polymerase chain reaction(RT-PCR). The expressions of VEGF in bone marrownbsp; fromnbsp; 20 cases of aplastic anemia and 20 normal controls were also determinednbsp; by immunohistochemistry assay.
