Methods RP-HPLC to Hypersil GOLD column (5μm 250mm × 4.60mm) for the analytical column, acetonitrile: water (plus phosphoric acid pH adjusted to 2.6) (eight ninety-two ~ 30:70) as mobile phase, flow rate: 0.8ml/min; detection wavelength was 254nm, column temperature 30 ℃. Results rutin concentration (0.0095 ~ 0.3800) μg within the range of chromatographic peak area showed good linearity. Conclusion The method is simple and accurate, reproducible, in the chromatographic conditions and sample handling method, can be accurately measured out the 20 species of rutin in Chinese Herbal Pieces. Keywords: Chinese medicine; rutin; high-performance liquid chromatography 【Abstract】 Objective To establish HPLC method for determination of the content of rutin in 20 kinds of Chinese herbal medicines.Methods The HPLC was conducted on a Hypersil GOLD column (5μm, 250mm × 4.6mm) with acetonitrile-water solution (8:92 ~ 30:70). The solution was adjusted to pH 2.6 with phosphoric acid and used as mobile phase with the flow rate of 0.8ml/min. The detective wavelength was set to 254 nm and column temperature at 30 ℃. Results The area under the peak of chromatography is very well correlated with the concentration of rutin in the range of (0.0095 ~ 0.3800) μg.Conclusion This method is simple and accurate. The experimental results are very reproducible. Under the conditions as stated in the method section, the rutin can be separated very well from 20 kinds of Chinese herbal medicines.
