 PAGE \* MERGEFORMAT 15 Interferon-α 2b right HL 60 cells in suppressing proliferation and promoting apoptosis 【Abstract】 In order to study IFN-α  2b (IFNα  2b) of the HL  60 cell proliferation and apoptosis, using Wright’s staining of HL  60 cells, morphological changes, acridine orange / ethidium bromide double staining observation of apoptosis, determined by observation of CD13 antigen expression and mature cell differentiation, MTT proliferative activity of experimental observation. The results showed that: by adding IFNα  2b 48 hours later, HL  60 cells increased apoptosis rate, proliferation activity declined, and along with IFNα  2b concentration increased, the inhibition become more pronounced. CD13 expression gradually decreased, the cells gradually mature into shape. Conclusion: IFNα  2b can promote apoptosis in HL  60 cells, inhibit HL  60 cells proliferation and promote maturation. The morphologic changes were observed by Wright ‘s and acridine orange (AO) and ethidium bromide (EB) staining respectively. Inhibition of proliferation was detected by MTT. Expression of CD13 was checked by indirect fluoroimmunoassay. The results showed that apoptosis rate of HL  60 cells assayed by the above  mentioned two methods was (51 ± 2)% and (78 ± 3)% respectively and OD570 values of proliferation inhibited were 1.8 ± 0.1 and 1.0 ± 0.1 respectively when the concentrations of the IFNα  2b were 500 and 10 000 U / ml in culture for 48 hours.
