Binding toanImportin-a molecule inthe cytoplasm is often requiredto mediatenuclear entry of a signaling protein. As multiple isoforms of Importin-a exist, some may be responsible for the entry of distinct cargoes rather than general nuclear import. Indeed, in neuronal systems, Importin-a isoforms can mediate very specific processessuchasaxonaltilingandcommunicationofaninjurysignal.Tostudynuclearimportduringdevelopment,we examinedtheexpressionandfunctionofImportin-a2inDrosophilamelanogaster.WefoundthatImportin-a2wasexpressed inthenervoussystemwhereitwasrequiredfornormalactivezonedensityattheNMJandaxonalcommissureformationin thecentralnervoussystem.OtheraspectsofsynapticmorphologyattheNMJandthelocalizationofothersynapticmarkers appeared normal in importin-a2 mutants. Importin-a2 also functioned in development of the body wall musculature. Mutantsinimportin-a2exhibitederrorsinmusclepatterningandorganizationthatcouldbealleviatedbyrestoringmuscle expressionofImportin-a2.Thus,Importin-a2isneededforsomeprocessesinthedevelopmentofboththenervoussystem andthelarvalmusculature.
