We show that saliva secretionwasgraduallyreducedto50%ofitsinitiallevel12weekspost-IR.Salivaproteincompositionwasfurtherexamined byproteomicanalysisfollowingmassspectrometry(MS)analysisthatrevealedproteinswithreducedexpressionoriginating from SSGs and proteins with increased expression derived from the serum, both indicating salivary tissue damage. To examinealterationsinmRNAexpressionlevelsmicroarrayanalysiswasperformed.Wefoundsignificantalterationsin95 genes,includingcell-cyclearrestgenes,SGfunctionalgenesandaDNArepairgene. Tissuedamagewasseenbyconfocal immunofluorescenceofa-amylaseandc-Kitthatshowedanincreaseanddecrease,respectively,inproteinexpression.This wascoherentwithreal-timePCRresults. ThisdataindicatesthatIRdamagestheSSGcells’abilitytoproduceandsecrete salivaandproteins,andmaintainthephysiologicalbarrierbetweenserumandsaliva.Thedamagedoesnothealduetocell- cyclearrest,whichpreventstissueregeneration.Takentogether,ourresultsrevealanewinsightintoIRpathobiology.
