(H.villosa)byaSingleDNAProbe ValentinaGrosso1,AnnaFarina1,AndreaGennaro2,DeboraGiorgi1,SergioLucretti1* 1ENEA,CASACCIAResearchCenter,Rome,Italy,2DepartmentofAgriculture,Forestry,NatureandEnergy-DAFNE,UniversityofTuscia,Viterbo,Italy Abstract Dasypyrumvillosum(L.)Candargy(sin.Haynaldiavillosa)isanannualwilddiploidgrassspecies(2n=2x=14;genomeVV) belongingtothePoaceaefamily,whichisconsideredtobeanimportantsourceofbioticandabioticstressresistancegenes forwheatbreeding.EnhancedcharacterizationofD.villosumchromosomescanfacilitateexploitationofitsgenepooland itsuseinwheatbreedingprograms.HerewepresentthecytogeneticidentificationofD.villosumchromosomesonslideby fluorescentinsituhybridization(FISH),withtheGAAsimplesequencerepeat(SSR)asaprobe.Wealsodescribetheisolation and the flow cytometric analysis of D. villosum chromosomes in suspension, resulting in adistinguished flow karyotype. Chromosomeswereflowsortedintothreefractions,accordingtheirDNAcontent,oneofwhichwascomposedofasingle type of chromosome, namely 6V, sorted with over 85% purity. Chromosome 6V is known to carry genes to code for importantresistanceandseedstoragecharacteristics,anditsisolationrepresentsanewsourceofgenetictraitsandspecific markersusefulforwheatimprovement.
