MelanosomalDynamicsAssessedwithaLive-Cell FluorescentMelanosomalMarker JanM.Bruder¤,ZacharyA.Pfeiffer,JonathanM.Ciriello,DianaM.Horrigan,NadineL.Wicks, BenjaminFlaherty,ElenaOancea* DepartmentofMolecularPharmacology,PhysiologyandBiotechnology,BrownUniversity,Providence,RhodeIsland,UnitedStatesofAmerica Abstract Melanocytespresentinskinandotherorganssynthesizeandstoremelaninpigmentwithinmembrane-delimitedorganelles calledmelanosomes.Exposureofhumanskintoultravioletradiation(UV)stimulatesmelaninproductioninmelanosomes, followedbytransferofmelanosomesfrommelanocytestoneighboringkeratinocytes.Melanosomalfunctioniscriticalfor protecting skin against UV radiation, but the mechanisms underlying melanosomal movement and transfer are not well understood.Herewereportanovelfluorescentmelanosomalmarker,whichweusedtomeasurereal-timemelanosomal dynamicsinlivehumanepidermalmelanocytes(HEMs)andtransferinmelanocyte-keratinocyteco-cultures.Afluorescent fusionproteinofOcularAlbinism1(OA1)localizedtomelanosomesinbothB16-F1cellsandHEMs,anditsexpressiondid not significantly alter melanosomal distribution. Live-cell tracking of OA1-GFP-tagged melanosomes revealed a bimodal kineticprofile,withmelanosomesexhibitingcombinationsofslowandfastmovement.WealsofoundthatexposuretoUV radiationincreasedthefractionofmelanosomesexhibitingfastversusslowmovement.Inaddition,usingOA1-GFPinlive co-cultures,wemonitoredmelanosomaltransferusingtime-lapsemicroscopy.TheseresultshighlightOA1-GFPasaspecific andeffectivemelanosomalmarkerforlive-cellstudies,revealnewaspectsofmelanosomaldynamicsandtransfer,andare relevanttounderstandingtheskin’sphysiologicalresponsetoUVradiation.
