Thecomplexityofglaucomahasmadeitdifficulttomodelinanimals,butalsochallengingtoidentifyresponsiblegenes.We haveusedzebrafishtoidentifyageneticallycomplex,recessivemutantthatshowsriskfactorsforglaucomaincludingadult onsetseveremyopia,elevatedIOP,andprogressiveretinalganglioncellpathology.Positionalcloningandanalysisofanon- complementingalleleindicatedthatnon-sensemutationsinlowdensitylipoproteinreceptor-relatedprotein2(lrp2 )underlie themutantphenotype.Lrp2,previouslynamedMegalin,functionsasanendocyticreceptorforawide-varietyofbioactive moleculesincludingSonichedgehog,Bonemorphogenicprotein4,retinol-bindingprotein,vitaminD-bindingprotein,and apolipoproteinE,amongothers.Detailedphenotypeanalysesindicatedthataslrp2mut
