Methods:Thestudypopulationconsistedofatotalof60patientswithseveresepsis,15withgramnegative bacteraemia,10healthycontrolsand60patientsundergoingelectivelungresectionsurgery.Pneumoniawas diagnosedbyCDCNNICcriteria.Geneexpressioninperipheralbloodleukocytes(PBLs)ofinterleukin(IL)-2,7,15 andinterferon(IFN)-g,Bax,Bim,Bcl-2wasdeterminedbyqRT-PCRandIL-2andIL-7serumproteinlevelsbyELISA. GeneexpressionofIL-2,7andIFN-gwasmeasuredinperipheralbloodleukocytes(PBL),culturedinthepresence oflipopolysacharide(LPS)andCD3bindingantibody(CD3ab) Results:IL-2geneexpressionwaslowerinthebacteraemiagroupcomparedwithcontrols,andlowerstillinthe sepsisgroup(P0.0001).IL-7geneexpressionwassimilarincontrolsandbacteraemia,butlowerinsepsis(P 0.0001).IL-15geneexpressionwassimilarinthethreegroups.Bcl-2geneexpressionwasless(P0.0001)andBim geneexpressionwasgreater(P=0.0003)inseveresepsiscomparedtobacteraemicandhealthycontrols.Baxgene expressionwassimilarinthethreegroups. Inlungresectionsurgerypatients,post-operativepneumoniawasassociatedwithaperioperativedecreaseinIL-2 mRNA(P0.0001)andIL-7mRNA(P=0.003).IL-2proteinlevelswerereducedinsepsisandbacteraemia comparedtocontrols(P=0.02)butsimilarinpneumoniaandnon-pneumoniagroups.IL-7proteinlevelswere similarinallgroups.
