PSIntegrinsandLaminins:KeyRegulatorsofCell MigrationduringDrosophilaEmbryogenesis JoseM.Urbano1¤,PalomaDom?′nguez-Gime′nez2,BeatrizEstrada1,Mar?′aD.Mart?′n-Bermudo1* 1CentroAndaluzdeBiolog?′adelDesarrollo,(CSIC)-UniversidadPablodeOlavide,Sevilla,Spain,2CentroAndaluzdeBiolog?′aMolecularyMedicinaRegenerativa,(CSIC)- UniversidaddeSevilla,Sevilla,Spain Abstract Duringembryonicdevelopment,therearenumerouscaseswhereorganortissueformationdependsuponthemigrationof primordialcells.IntheDrosophilaembryo,thevisceralmesoderm(vm)actsasasubstrateforthemigrationofseveralcell populations ofepithelialorigin,includingtheendoderm,thetracheaandthesalivary glands.Thesemigratoryprocesses requirebothintegrinsandlaminins.ThecurrentmodelisthataPS1bPS(PS1)and/oraPS3bPS(PS3)integrinsarerequiredin migratingcells,whereasaPS2bPS(PS2)integrinisrequiredinthevm,whereitperformsanasyetunidentifiedfunction. Here,weshowthatPS1integrinsarealsorequiredforthemigrationoverthevmofcellsofmesodermalorigin,thecaudal visceralmesoderm(CVM).TheseresultssupportamodelinwhichPS1mighthaveevolvedtoacquirethemigratoryfunction ofintegrins,irrespectiveoftheoriginofthetissue.Thisintegrinfunctionishighlyspecificanditsspecificityresidesmainlyin the extracellular domain. In addition, we have identified the Laminin a1,2 trimer, as the key extracellular matrix (ECM) componentregulatingCVMmigration.Furthermore,weshowthat,asitisthecaseinvertebrates,integrins,andspecifically PS2,contributestoCVMmovementbyparticipatinginthecorrectassemblyoftheECMthatservesastracksformigration.
