Methods:One-hundredandthirtyHCCpatientswhohadundergonecurativeliverresectionwereselectedand immunohistochemistry,Westernblotting,andquantitativerealtimepolymerasechainreaction(Q-PCR)were performedtoanalyzeSOX9expressionintherespectivetumors. Results:Immunohistochemistry,Westernblotting,andQ-PCRconsistentlyconfirmedSOX9overexpressioninHCC tissuescomparedwiththeiradjacentnonneoplastictissues(P0.01).Additionally,immunostainingshowedmore SOX9positivecellsinthehighertumorstage(T3~4)andtumorgrade(G3)thaninthelowertumorstage(T1~2, P=0.03)andtumorgrade(G1~2,P=0.01),respectively.Moreover,HCCpatientswithhighSOX9expressionwere significantlyassociatedwithlower5-yearoverallsurvival(P0.01)andlower5-yeardisease-freesurvival(P0.01), respectively.TheCoxproportionalhazardsmodelfurthershowedthatSOX9over-expressionwasanindependent poorprognosticfactorforboth5-yeardisease-freesurvival(hazardsratio[HR]=2.621,95%confidenceinterval [CI]=1.548-5.829,P=0.01)and5-yearoverallsurvival(HR=3.825,CI=1.638-7.612,P=0.003)inHCC. Conclusion:OurdatasuggestforthefirsttimethattheoverexpressionofSOX9proteininHCCtissuesisof predictivevalueontumorprogressionandpoorprognosis.
