國立屏東科技大學食品科學系 題目: Repair effect of onion (Allium cepa L.) extract on alcohol-induced injury of liver cell 研究生預口試摘要 指導教授: 老師吳明昌報告:陳宛伶 報告日期:.04.23 摘要 酒有特的風味,含有維生素、等質,攝取過多的酒精會造成肝細胞損傷,進一步導致動脈粥狀硬化症、脂肪肝、肝纖維化及誘發肝癌等。槲皮素類黃酮,能夠螯金屬離子、清除氧自由基具有抗氧化的特性,研究顯示洋蔥所萃取出之槲皮素,具有、抗過敏及預防心血管病變等功效,對健康有益。本研究利用洋蔥萃取液,肝細胞酒精損傷之。結果顯示,利用0.5濃度之酒精培養6小時,誘導FL83B細胞,,WST-1、ALTAlanine aminotransferase)、GGT-glutamyl transferase)的測試觀察細胞受損的情形noids, total phenolic, total anthocyanin等的多酚含量檢驗,結果顯示,以紅洋蔥所含萃取出的多酚含量較多,而抗氧化的效果也較佳,由上述結論推測,FLB3B細胞的修復效果與抗氧化作用有相關性。關鍵字: Abstract The wine contains special flavor and some nutrients such as vitamins and organic acids. Intake more alcohol would lead to injure the liver cell, such as atherosclerotic, fibrosis, liver cancer etc. Quercetin is one of the flavonoids wildly distributed in the nature and showed the chelating metallic ion, scavenging free radical on the antioxidant characterization, several reports showed the extract flavonoids of Allium cepa L. can inhibit caner, anti-virus, anti-allergic inflammation and reduce the blood fats etc., it is beneficial to the human health. The Allium cepa L. extract on repair of ethanol damage liver cell will be investigated in this study. The sample from different variety (red, yellow, white), drying treatment (freezing, 60℃ hot-air), extraction solvent (70% alcohol, distiller water) and different concentration (0.25, 0.5, 1, 2) were treated on the cell, and FL83B cultivated with alcohol for 6 hours with the concentration of 0.25M has been used as the alcohol injury cell model. 70% alcohol extraction of red Allium cepa L.with freezing drying was added to cell on 24 hour showed the best on cell repair by the WST-1, ALT (Alanine aminotransferase), GGT (γ-glutamyl transferase) tests to the damaged liver cells. The flavonoids, total phenolic, total anthocyanin assay and the antioxidant assay by TEAC, DPPH to Allium cepa L. also were executed, the results showed there was much total phenolic and better in antioxidant ability from the
