结直肠癌中抑癌基因ING1的表达研究 作者：张森 韦建宝 周永淳 陈利生 【摘要】 目的 探讨抑癌基因ING1及其蛋白p33/ING1在结直肠癌中的表达。 方法 用RTPCR检测35例散发性结直肠癌中ING1 mRNA的表达水平，并 应用 SP法检测60例散发性结直肠癌及正常黏膜组织中p33/ING1蛋白的表达。结果 结直肠癌组织、正常黏膜组织中p33/ING1蛋白阳性表达率分别为43.3%(26/60)、100%(60/60)，两者比较差异有统计学意义(P0.01)。p33/ING1在无淋巴结转移组及淋巴结转移组中的阳性表达率分别为57.6%(19/33)、25.9%(7/27)，两者比较差异有统计学意义(P0.05)。在Dukes A、B期和Dukes C、D期患者癌组织中p33/ING1的阳性表达率分别为56.7%(17/30)、30.0%(9/30)，两者比较差异有统计学意义(P0.05)。结论 ING1及p33/ING1蛋白的低表达与散发性结直肠癌发生、 发展 密切相关，临床分期越差者表达水平越低。 【关键词】 抑癌基因 ING1 结肠肿瘤 直肠肿瘤 【Abstract】 Objective To discuss the expression of tumor suppressor gene ING1 and its protEin p33/ING1 in development of sporadic colorectal carcinoma. Methods Reverse transcriptionpolymerase chain reaction was used to detect mRNA expression level of ING1 in 35 cases with sporadic colorectal carcinoma and streptomycete avidinperoxidase immunohistochemistry to measure expression level of protEIn p33/ING1 in colorectal cancerous tissues and normal mucous tissues from 60 cases with sporadic colorectal carcinoma. Results Expression level of p33/ING1 mRNA in colorectal cancerous tissues was more significantly decreased than that in normal mucous tissues. Expression level of p33/ING1 mRNA in the tumors of patients at stage of Dukes A and B was significantly higher than that in patients at stage of Dukes C and D, as indicated that the expression decreased with poor clinical stage. Positive expression rates of p33/ING1 in colorectal cancerous tissues and normal mucous tissues were 43.3% (26/60) and 100% (60/60), respectively, with significant statistical difference (P0.01). Positive expression rates of p33/ING1 in groups with or without lymph node metastasis were 25.9% (7/27) and 57.6% (19/33) respectively (P0.05). Positive expression rates of p33/ING1 in patients at stage of Dukes A, B and at stage of Dukes C and D were 56.7% (17/30) and 30.0% (9/30) respectively (P0.05).
