The lowest bacteriostatic concentration of antibiotics (MIC) and half bacteriostasis (MIC) method
Several methods for determining the lowest concentrations of antimicrobial agents (MIC) :
Constant broth dilution
1.1.1. The concentration of antimicrobial drug storage liquid should not be less than 1000 mu g/ml (such as 1280 mu g/ml) or 10 times the highest concentration. Antibacterial drugs with low solubility can be slightly lower than these concentrations. Antibacterial drugs are purchased directly from manufacturers or related agencies. The formula of solution quantity or powder dosage of antimicrobial agent is calculated. For example, you need to make a 100 ml concentration of antibiotics for 1, 1280 mu g/ml, and the antibiotic is a powder, and the drug has a potency of 750 mu g/mg. Using analytical scales, the amount of an antibiotic powder was 182.6 mg. According to the formula, the dosage of the required diluent is: (182.6 m g x 750 mu g/ml) / 1280 mu g/ml = 107.0 ml, and then the 182.6 mg of antibiotic powder dissolved in 107.0 ml diluents. The solvents and thinners used in the preparation of antimicrobial drug storage are shown in table 5. Make good antibacterial drug storage liquid should be stored at 60 ℃ the following environment, no more than 6 months shelf life.
1.1.2. The drug sensitive test concentration range with antibacterial drugs Antibacterial drug sensitivity test according to NCCLS standard operation, drug concentration range should contain resistant, mediation, and sensitive to the boundary value, special exception.
1.1.3. The medium NCCLS recommended using Mueller - Hinton (MH) broth, pH7.2 ~ 7.4. Aerobic and facultative anaerobic bacteria grow well in this medium.
