Whole-cell, ins ide-out and on-cell mode patch clam p techniqueswere used to record m embrane currents after dialyzing the in tracellular Ca2+ or applying the calm odu lin inh ibitor, W-7. R esults A lmost all the cultured cellsw ere mi munopos itive forC-K it an tibody protein. Inw hole-cell patch clamp m ode, decreasing the intracellular free Ca2+ by using 10 mm ol/L EGTA in pipette, sustained inw ard hold ing currentsw ere activated in ICC, wh ile no response in another ICC. W-7 elicited sustained inw ard holding current and potentiated the am plitude of spontaneous transient inw ard currents in ICC, w hile no response in another ICC. In inside-out patch clamp m ode, when the intracellular calcium w as decreased from 1m ol/L to 0. 1 m ol/L, s ingle channel activitiesw ere sign ifican tly potentiated.
