Sample preparation methods and the factors affecting the AFM image of polysaccharide are described in detail. Advance in using AFM for morphological observation and quantitative study on polysaccharide molecules are reviewed. Research work on single molecular mechanical spectroscopy of polysaccharide and determination and adjustment of conformational change of sugar residue are introduced. Prospects for the further application of AFM in polysaccharide investigation is presented. Key words: Atomic force microscope; Polysaccharide 1982 ·
