Characteristics of agarose gel
Natural agar (agar) is a polysaccharide consisting mainly of agarose (agarose, about 80%) and agar gel (agaropectin). Agarose is composed of galactose neutral compounds and its derivatives, without charge, and agar gel is a sulfate containing carboxyl and strong acidic polysaccharides, because these groups with charge, can produce strong electroosmosis in the electric field, and the sulfate and some egg white matter the effect of electrophoresis rate and the separation effect. Therefore, the current use of agarose as an electrophoretic support for slab electrophoresis has the following advantages.
(1) agarose gel electrophoresis is simple and fast, and the sample can be electrophoresis without pretreatment.
(2) agarose gel structure uniform, water (about 98%~99%), approximate free electrophoresis samples than the free diffusion current, the sample adsorption is minimal, so clear electrophoretogram, high resolution and good repeatability.
(3) transparent and non violet absorption of agarose. The electrophoresis process and results can be directly detected and quantified by ultraviolet lamp.
(4) the electrophoresis zone is easy to dye, and the sample is easy to elution, so it is easy to determine quantitatively. Made of dry film, can be preserved for a long time.
Currently, agarose is often used as an electrophoretic support to separate proteins and isoenzymes. The combination of agarose electrophoresis with immunochemical development into immunoelectrophoresis can identify complex systems that cannot be identified by other methods. 0.1ug protein can be detected by the establishment of ultramicro technology.
Agarose gel electrophoresis is also used to isolate and identify nucleic acids, such as DNA identification and restriction endonuclease mapping of DNA. Because this method is easy to operate, simple in equipment, requiring few samples and high resolving power, it has become one of the most common experimental methods
