Sample pre-treatment and method validation 样品前处理与测定方法的效能指标 基本要求 掌握氧瓶燃烧法的基本原理、吸收液的选择原则； 熟悉不经有机破坏的分析方法和湿法破坏、干法破坏； 掌握测定方法的精密度、准确度、检测限、定量限、选择性、线性与范围、重现性和耐用性等内容。 Fundamental requirement Mastering the basic principle and the selection of absorbing solution in oxygen flask combustion ； Familiarizing with the analytical methods without organic destruction、wet and dry destruction methods； Mastering the method validation items: precision、accuracy、detection limit、quantitative limit、selectivity、linearity 、range、repeatability、robustness of determination methods. Pre-treatment of qualitative analysis and typical validation characteristics 定量分析样品的前处理方法 不经有机破坏的分析方法： ①直接测定法； ②经水解后测定法：直接回流滴定法 、用硫酸水解后测定法； ③经氧化还原后测定法：碱性还原后测 定、酸性还原后测定、汞齐化法、利用药物中可游离的金属离子的氧化性测定含量。 有机破坏后的分析方法 ： Pre-treatment of qualitative analysis Analytical methods without organic destruction： ①Direct determination； ②Determination after hydrolyzing：direct reflux determination 、determination after hydrolyzing with sulfuric acid ； ③Redox determination：determination after base or acid reduction 、 amalgamation method、content determination using the oxidation of free metallic ions in drugs. Analytical method with organic destruction: （一）湿法破坏：硝酸-高氯酸法、硝酸-硫 酸法、硫酸-硫酸盐法、其他湿法； （二）干法破坏：本法适用于湿法不易破坏完全的有机物以及某些不能用硫酸进行破坏的有机药物。不适用于含易挥发性金属（如汞、砷等）有机药物的破坏。 （三）氧瓶燃烧法：系将有机药物放入充满氧气的密闭的燃烧瓶中进行燃烧，并将燃烧所产生的欲测物质吸收于适当的吸收液中，然后根据欲测物质的性质，采用适宜的分析方法进行鉴别、检查或测定含卤素有机药物或含硫、氮、硒等其它元素的有机药物。 （一）Wet destruction：nitric acid- perchloric acid method、nitric acid-sulfuric acid method、sulfuric acid-sulfate method、and other wet methods； （二）Dry destruction：This method is used for organic substances which are not easily destroyed by wet method and some organic drugs which can not be destroyed by sulfuric acid. Organic drugs containing easily volatile metals, such as mercury and arsenic, is not suitable. Oxygen flask combustion
