宫颈上皮内瘤变和宫颈癌组织中Survivin和Bcl-2表达与相关性.docVIP

【摘要】 　　目的 ：研究Survivin、Bcl-2在宫颈上皮内瘤变及宫颈癌中的表达，探讨两者在宫颈癌发病机制中的作用。方法：采用原位杂交法检测宫颈上皮内瘤变（CIN）和宫颈癌中Survivin基因的表达，以正常宫颈组织为对照。同时采用免疫组织化学法检测Bcl-2蛋白在各组中的表达。结果 ：Survivin mRNA的阳性率在宫颈癌、CIN和正常宫颈组织中分别为77.8%、44.6%和0%，Bcl-2蛋白在三者中的阳性表达率分别为70.4%、39.2%和0，两者差异均有显著性。Survivin及Bcl-2阳性表达在CINⅢ级中明显高于CIN I/Ⅱ级，在宫颈癌Ⅱb～Ⅲ期中显著高于Ⅰ～Ⅱa期，在低分化组中显著高于高、中分化组。在淋巴结转移组中Survivin的表达显著高于无淋巴结转移组。在宫颈癌中Survivin与Bcl-2的表达呈正相关。结论：Survivin和Bcl-2的异常表达可能参与宫颈癌的发生、发展过程，可能有协同促进抗凋亡作用，并提示与肿瘤的恶性程度和不良预后有关。 【关键词】 Survivin Bcl-2 宫颈癌 CIN 　　Expression and correlation between Survivin and Bcl-2 in cervical intraepithelial neoplasia and cervical carcinoma 　　ZHU Hua，SHI Zhengzheng，ZHANG Huxiang，WAN Li，YANG Xiaojun，ZHENG Feiyun. 　　Department of Gynecology and Obstetrics，the First Affiliated Hospital of Wenzhou Medical College，Wenzhou，325000 　　Abstract: Objective: To investigate the expression of Survivin and Bcl-2 in cervical intraepithelial neoplasia and cervical carcinoma， and to explore the role of them in cervical carcinoma pathogenesis. Methods: The expression of Survivin mRNA was detected with hybridiza-tion in situ in CIN and cervical carcionma，and the expression of Bcl-2 was detected with immunohistochemical method. Normal cervical tissues were acted as the control. Results: The positive rates of Survivin mRNA in cervical carcinoma，CIN and the normal cervical tissues were 77.8%，44.6% and 0%，respectively；while the expression of Bcl-2 was 70.4%，39.2% and 0% in the above three groups. There was significant difference among those groups. The expression of Survivin and Bcl-2 in CINⅢ was obviously more significant than those in CIN I/Ⅱ. And the differences were obviously more significant in cervical carcinoma at stage Ⅱb～Ⅲ than those at stageⅠ～Ⅱa. The expressions of Survivin and Bcl-2 with poorly differentiation were also obviously more significantly different than those with good or moderated differentiation.
