火把花根片对EAE大鼠脑及脊髓L-选择素表达影响 摘 要：目的：探讨火把化根片对实验性自身免疫性脑脊髓炎(EAE)大鼠的治疗作用及L-选择素的影响。方法:模型组采用豚鼠髓鞘蛋白匀浆和福氏完全佐剂诱发大鼠急性EAE；治疗组在模型组基础上予以火把化根(600mg·kg-1·d-1)治疗，观察临床表现并评分；HE染色和Loyez氏髓鞘染色观察病理和髓鞘改变；免疫组化技术测定脑和脊髓的L-选择素水平。结果:治疗组未出现临床症状，L-选择素为1.88±0.64，与模型组比较P0.05；脑和脊髓小血管周围炎症细胞浸润明显减少；髓鞘结构完整。结论：火把化根片对EAE有治疗作用，其机制与抑制L-选择素表达有关。 关键词：实验性自身免疫性脑脊髓炎；火把化根；L-选择素 中图分类号：R285.5文献标识码：A文章编号：1673-7717(2011)12-2744-02 Influence of L-selectin on Experimental Allergic Encephalomyelitis of Wistar Rats by the Treatment of Colquhoumai Root Tablet ZHENG Hezhong，XU Kewen，DONG Wei，WANG Yulan，NI Qin，WENG Jing，YI Hong (Department of Neurology, The People’s Hospital of Yuyao City,Yuyao 315400，Zhejiang，China) Abstract:Objective：To investigate the influence of L-selectin on experimental allergic encephalomyelitis (EAE) of Wistar rats by the treatment of Colquhoumai root tablet. Methods：The animal models were established in rats by immunization with myelin basic protein of spinal cord of guinea pig and complete Freund’s adjuvant. On the base of the animal models, Colquhoumai root tablet was administered with dose(600mg.kg-1.d-1) in treatment groups. The symptom of EAE was observed; pathological feature and myelin of brain and spinal were detected with HE stain and Loyez’s stain respectively. The levels of L-selectin were detected by immunohistochemistry. Results：None developed EAE in therapy groups.The levels of L-selectin was 1.88±0.64, obviously lower than that of model groups (P0.05).Inflammation cells which around vessels and demyelization in white matter of brain and spinal cords were reduced. Conclusions：Colquhoumai root tablet can treat EAE.Mechanisms of Colquhoumai root tablet on EAE may be correlated with its ability to depress the expression of L-selectin.
